This frequency is not significantly higher than expected for a pathogenic variant in LPL causing Familial Lipoprotein Lipase Deficiency (0.0001 vs 0.0034), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.8G>A in individuals affected with Familial Lipoprotein Lipase Deficiency and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. The serine at codon 3 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
