In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This sequence change replaces tryptophan, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 506 of the CHRNA2 protein (p.Trp506Arg). This variant is present in population databases (no rsID available, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with CHRNA2-related conditions. This missense change has been observed in at least one individual who was not affected with CHRNA2-related conditions (Invitae). ClinVar contains an entry for this variant (Variation ID: 543534).
