The c.2360T>G (p.L787R) alteration is located in exon 20 (coding exon 19) of the WRN gene. This alteration results from a T to G substitution at nucleotide position 2360, causing the leucine (L) at amino acid position 787 to be replaced by an arginine (R). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
Werner syndrome Uncertain:1
This sequence change replaces leucine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 787 of the WRN protein (p.Leu787Arg). This variant is present in population databases (no rsID available, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with WRN-related conditions. ClinVar contains an entry for this variant (Variation ID: 458416). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. RNA analysis performed to evaluate the impact of this missense change on mRNA splicing indicates it does not significantly alter splicing (Invitae).
