The c.3340C>T (p.P1114S) alteration is located in exon 28 (coding exon 27) of the WRN gene. This alteration results from a C to T substitution at nucleotide position 3340, causing the proline (P) at amino acid position 1114 to be replaced by a serine (S). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces proline, which is neutral and non-polar, with serine, which is neutral and polar, at codon 1114 of the WRN protein (p.Pro1114Ser). This variant is present in population databases (rs749621372, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with WRN-related conditions. ClinVar contains an entry for this variant (Variation ID: 570858). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. RNA analysis performed to evaluate the impact of this missense change on mRNA splicing indicates it does not significantly alter splicing (Invitae).
