Inborn genetic diseases Uncertain:1
The c.115C>T (p.H39Y) alteration is located in exon 2 (coding exon 2) of the ADAM9 gene. This alteration results from a C to T substitution at nucleotide position 115, causing the histidine (H) at amino acid position 39 to be replaced by a tyrosine (Y). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
not provided Uncertain:1
This sequence change replaces histidine, which is basic and polar, with tyrosine, which is neutral and polar, at codon 39 of the ADAM9 protein (p.His39Tyr). This variant is present in population databases (rs768310031, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with ADAM9-related conditions. ClinVar contains an entry for this variant (Variation ID: 1514827). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
