This sequence change replaces proline, which is neutral and non-polar, with histidine, which is basic and polar, at codon 401 of the NBN protein (p.Pro401His). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with NBN-related conditions. ClinVar contains an entry for this variant (Variation ID: 219698). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. This variant is denoted NBN c.1202C>A at the cDNA level, p.Pro401His (P401H) at the protein level, and results in the change of a Proline to a Histidine (CCC>CAC). This variant has not, to our knowledge, been published in the literature as a pathogenic or benign germline variant. NBN Pro401His was not observed in large population cohorts (Lek 2016). This variant is not located in a known functional domain. In silico analysis, which includes protein predictors and evolutionary conservation, supports that this variant does not alter protein structure/function. Based on currently available evidence, it is unclear whether NBN Pro401His is a pathogenic or benign variant. We consider it to be a variant of uncertain significance. -

The p.P401H variant (also known as c.1202C>A), located in coding exon 10 of the NBN gene, results from a C to A substitution at nucleotide position 1202. The proline at codon 401 is replaced by histidine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
