The encoded protein is a member of the MRE11/RAD50 double-strand break repair complex which consists of 5 proteins. This gene product is thought to be involved in DNA double-strand break repair and DNA damage-induced checkpoint activation. [provided by RefSeq, Jul 2008]

The glutamic acid at codon 259 is replaced by lysine, an amino acid with similar properties. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, c.775G>A has not been reported in the literature in individuals affected with Nijmegen Breakage Syndrome, and no experimental evidence demonstrating an impact on protein function has been published. Three clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All laboratories classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance. -
