The p.L165I variant (also known as c.493C>A), located in coding exon 5 of the NBN gene, results from a C to A substitution at nucleotide position 493. The leucine at codon 165 is replaced by isoleucine, an amino acid with highly similar properties. This variant was reported in 1/60,466 breast cancer cases and in 2/53,461 controls (Dorling et al. N Engl J Med. 2021 02;384:428-439). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
