Variant summary: TMEM67 c.1351C>T (p.Arg451X) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. The variant allele was found at a frequency of 8e-06 in 251374 control chromosomes (gnomAD). c.1351C>T has been reported in the literature in multiple individuals affected with Joubert syndrome, Meckel-Gruber syndrome and COACH syndrome (e.g. Consugar_2007, Doherty_2010, Chaki_2011). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Three submitters have provided assessments for this variant to ClinVar after 2014 and all classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -

This premature translational stop signal has been observed in individual(s) with Joubert syndrome (PMID: 17377820, 21866095, 28497568). ClinVar contains an entry for this variant (Variation ID: 217724). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. -
