It had not been previously curated by ICSL or reported in the Human Gene Mutation Database (HGMD: prior to June 1st, 2018), and was therefore a candidate for classification through an automated scoring system. Utilizing variant allele frequency, disease prevalence and penetrance estimates, and inheritance mode, an automated score was calculated to assess if this variant is too frequent to cause the disease. Based on the score, this variant could not be ruled out of causing disease and therefore its association with disease required further investigation. A literature search was performed for the gene, cDNA change, and amino acid change (if applicable). No publications were found based on this search. This variant was therefore classified as a variant of unknown significance for this disease. -

The p.G125V variant (also known as c.374G>T), located in coding exon 4 of the FKTN gene, results from a G to T substitution at nucleotide position 374. The glycine at codon 125 is replaced by valine, an amino acid with dissimilar properties. This alteration has been reported in a sudden unexplained death cohort, an ocular disorders cohort, and a dilated cardiomyopathy (DCM) cohort (Lahrouchi N et al. Eur J Hum Genet, 2020 Jan;28:17-22; Mazzarotto F et al. Circulation, 2020 Feb;141:387-398; Deml B et al. Eur J Hum Genet, 2016 Apr;24:535-41). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
