Genes affected
WHRN (HGNC:16361): (whirlin) This gene is thought to function in the organization and stabilization of sterocilia elongation and actin cystoskeletal assembly, based on studies of the related mouse gene. Mutations in this gene have been associated with autosomal recessive non-syndromic deafness and Usher Syndrome. Alternative splicing results in multiple transcript variants. Uncertain significance, criteria provided, single submitterclinical testingLabcorp Genetics (formerly Invitae), LabcorpJun 13, 2022This sequence change replaces valine, which is neutral and non-polar, with isoleucine, which is neutral and non-polar, at codon 454 of the WHRN protein (p.Val454Ile). This variant is present in population databases (rs370178817, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with WHRN-related conditions. ClinVar contains an entry for this variant (Variation ID: 45651). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The isoleucine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

Likely benign, criteria provided, single submitterclinical testingLaboratory for Molecular Medicine, Mass General Brigham Personalized MedicineFeb 22, 2013Val454Ile in exon 6 of DFNB31: This variant has been identified in 0.01% (1/8600 ) of European American chromosomes in a broad population by the NHLBI Exome sequ encing project (http://evs.gs.washington.edu/EVS/). This variant is not expected to have clinical significance due to a lack of conservation across species, inc luding mammals. Of note, at least 8 different mammals (bushbaby, mouse, rat, kan garoo rat, guinea pig, tenrec, opossum and platypus) have an Isoleucine (Ile). I n addition, computational analyses (PolyPhen2, SIFT, AlignGVGD) suggest the vari ant will not impact the protein. -
