Variant summary: CRAT c.1811C>T (p.Ala604Val) results in a non-conservative amino acid change located in the Choline/carnitine acyltransferase domain (IPR039551) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 5.2e-05 in 250038 control chromosomes. To our knowledge, no occurrence of c.1811C>T in individuals affected with Neurodegeneration With Brain Iron Accumulation 8 and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 2419947). Based on the evidence outlined above, the variant was classified as uncertain significance. -
not provided Uncertain:1
This sequence change replaces alanine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 604 of the CRAT protein (p.Ala604Val). This variant is present in population databases (rs779910980, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with CRAT-related conditions. ClinVar contains an entry for this variant (Variation ID: 2419947). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt CRAT protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
