The p.H1053Q variant (also known as c.3159C>A), located in coding exon 21 of the TSC1 gene, results from a C to A substitution at nucleotide position 3159. The histidine at codon 1053 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
