This variant has been reported in a sample of the TCGA renal cell carcinomas (RCC) cohort from cbioportal (www.cbioportal.org) (PMID: 29941307). This variant is absent in the general population by the Genome Aggregation Database (gnomAD). Truncating variants in TSC1 are known to be pathogenic (PMID: 10363127, 10533067, 20399389). This variant has been reported in ClinVar (Variation ID: 1953076). Therefore, the c.1257dup (p.Arg420Glnfs*22) variant of the TSC1 gene is classified as pathogenic. -
