Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4.3e-06 in 231476 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.253G>T in individuals affected with Cutaneous Malignant Melanoma and no experimental evidence demonstrating its impact on protein function have been reported. Three clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014, and all laboratories classified the variant as uncertain significance. Additionally, variants occurring at the same nucleotide (c.253G>A, p.Ala85Thr and c.253G>C, p.Ala85Pro) and within the same codon (c.254C>T, p.Ala85Val) have all been reported as uncertain significance in ClinVar. This variant has not been reported in the literature in individuals affected with CDKN2A (p16INK4a)-related conditions.
