In a cell cycle activity assay, this alteration showed normal binding to CDK4 and CDK6 proteins; however, authors noted that normal binding to CDK4 or CDK6 is a poor predictor of cell cycle function and pathogenicity and a normal result in the assay does not change the odds that a variant is pathogenic or neutral (Miller PJ et al. Hum Mutat. 2011; 32:900-11). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
