DNAI1 (HGNC:2954): (dynein axonemal intermediate chain 1) This gene encodes a member of the dynein intermediate chain family. The encoded protein is part of the dynein complex in respiratory cilia. The inner- and outer-arm dyneins, which bridge between the doublet microtubules in axonemes, are the force-generating proteins responsible for the sliding movement in axonemes. The intermediate and light chains, thought to form the base of the dynein arm, help mediate attachment and may also participate in regulating dynein activity. Mutations in this gene result in abnormal ciliary ultrastructure and function associated with primary ciliary dyskinesia and Kartagener syndrome. This sequence change replaces glycine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 497 of the DNAI1 protein (p.Gly497Val). This missense change has been observed in individual(s) with primary ciliary dyskinesia (Invitae). For these reasons, this variant has been classified as Pathogenic. This variant disrupts the c.1490G nucleotide in the DNAI1 gene. Other variant(s) that disrupt this nucleotide have been determined to be pathogenic (PMID: 16858015). This suggests that this nucleotide is clinically significant, and that variants that disrupt this position are likely to be disease-causing. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant  is likely to be tolerated.
