Primary ciliary dyskinesia Uncertain:3
This sequence change replaces histidine with asparagine at codon 608 of the DNAI1 protein (p.His608Asn). The histidine residue is moderately conserved and there is a small physicochemical difference between histidine and asparagine. This variant is present in population databases (rs148762102, ExAC 0.002%). This variant has not been reported in the literature in individuals affected with DNAI1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The c.1822C>A (p.H608N) alteration is located in exon 19 (coding exon 19) of the DNAI1 gene. This alteration results from a C to A substitution at nucleotide position 1822, causing the histidine (H) at amino acid position 608 to be replaced by an asparagine (N). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
