GALT (HGNC:4135): (galactose-1-phosphate uridylyltransferase) Galactose-1-phosphate uridyl transferase (GALT) catalyzes the second step of the Leloir pathway of galactose metabolism, namely the conversion of UDP-glucose + galactose-1-phosphate to glucose-1-phosphate + UDP-galactose. The absence of this enzyme results in classic galactosemia in humans and can be fatal in the newborn period if lactose is not removed from the diet. The pathophysiology of galactosemia has not been clearly defined. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Apr 2012]

This variant is also absent from general population databases (1000 Genomes Project, Exome Variant Server, and Genome Aggregation Database), indicating it is not a common polymorphism. The asparagine at codon 97 is highly conserved, is involved in ligand binding (Boutron 2012), and computational analyses (SIFT, PolyPhen-2) predict that this variant is deleterious. Other variants at this codon (c.290A>G, p.Asn97Ser; c.289A>G; p.Asn97Asp) have been reported in individuals with galactosemia and are considered pathogenic (Boutron 2012, Shin 2004). Based on available information, the p.Asn97Ile variant is considered to be likely pathogenic. References: Boutron A et al.
