The variant was absent in 251484 control chromosomes (gnomAD). c.616C>T has been reported in the literature in individuals affected with Galactosemia, including one homozygote and one compound heterozygote with a second pathogenic allele (GALT c.584T>C [p.Leu195Pro] ClinVar:25222). These data indicate that the variant is associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
