The members of the Fanconi anemia complementation group do not share sequence similarity; they are related by their assembly into a common nuclear protein complex. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. This variant disrupts the the last 40 amino acid residue in FANCG. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 10567393, 10961856). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. ClinVar contains an entry for this variant (Variation ID: 188303). This premature translational stop signal has been observed in individual(s) with Fanconi anemia (PMID: 23613520).
