It is a rate-limiting enzyme in the sialic acid biosynthetic pathway. Sialic acid modification of cell surface molecules is crucial for their function in many biologic processes, including cell adhesion and signal transduction. Differential sialylation of cell surface molecules is also implicated in the tumorigenicity and metastatic behavior of malignant cells. Mutations in this gene are associated with sialuria, autosomal recessive inclusion body myopathy, and Nonaka myopathy. The variant allele was found at a frequency of 4e-06 in 251360 control chromosomes. c.479G>A has been reported in the literature in compound heterozygous individuals affected with Inclusion Body Myopathy 2 (Chaouch_2014, Granger_2022, Mori-Yoshimura_2012, Tomimitsu_2004, Guo_2022). These data indicate that the variant is very likely to be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in hyposialylation in human pluripotent stem cells (hPSCs) (Park_2022).
