Genes affected
TMC1 (HGNC:16513): (transmembrane channel like 1) This gene is considered a member of a gene family predicted to encode transmembrane proteins. The specific function of this gene is unknown; however, it is known to be required for normal function of cochlear hair cells. Mutations in this gene have been associated with progressive postlingual hearing loss and profound prelingual deafness. [provided by RefSeq, Jul 2008]

Pathogenic, criteria provided, single submitterclinical testingGeneDxMar 31, 2021Frameshift variant predicted to result in protein truncation or nonsense mediated decay in a gene for which loss-of-function is a known mechanism of disease; Not observed in large population cohorts (Lek et al., 2016); Has not been previously published as pathogenic or benign to our knowledge -
Rare genetic deafness Pathogenic:1
Pathogenic, criteria provided, single submitterclinical testingLaboratory for Molecular Medicine, Mass General Brigham Personalized MedicineJun 04, 2015The p.Met413fs variant in TMC1 has been previously reported in one individual wi th hearing loss by our laboratory who was compound heterozygous for this variant and a second truncating variant in TMC1 (LMM unpublished data). This variant is absent from large population studies. This frameshift variant is predicted to a lter the protein's amino acid sequence beginning at position 413 and lead to a p remature termination codon 4 amino acids downstream. This alteration is then pre dicted to lead to a truncated or absent protein. Loss of function variants affec ting both copies of TMC1 is an established disease mechanism for nonsyndromic au tosomal recessive hearing loss. In summary, this variant meets our criteria to b e classified as pathogenic for hearing loss in an autosomal recessive manner bas ed upon the predicted impact of the variant. -
