The variant was also identified in control databases in 180 of 282824 chromosomes (1 homozygous) at a frequency of 0.000636 increasing the likelihood this could be a low frequency benign variant (Genome Aggregation Database Feb 27, 2017). The variant was observed in the following populations: African in 166 of 24964 chromosomes (freq: 0.00665), Latino in 12 of 35434 chromosomes (freq: 0.000339), Other in 1 of 7224 chromosomes (freq: 0.000138) and European (non-Finnish) in 1 of 129142 chromosomes (freq: 0.000008), while the variant was not observed in the Ashkenazi Jewish, East Asian, European (Finnish), and South Asian populations. The p.Ile123 residue is not conserved in mammals and computational analyses (PolyPhen-2, SIFT, AlignGVGD, BLOSUM, MutationTaster) do not suggest a high likelihood of impact to the protein; however, this information is not predictive enough to rule out pathogenicity.
