The members of the Fanconi anemia complementation group do not share sequence similarity; they are related by their assembly into a common nuclear protein complex. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 7.9e-06 in 252656 control chromosomes (gnomAD and publication data). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.521+10_521+13delAAAC has been reported in the literature in two individuals affected with esophageal squamous cell carcinoma (Akbari_2011). The report does not provide unequivocal conclusions about association of the variant with Fanconi Anemia Group C. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. One ClinVar submitter (evaluation after 2014) cites the variant as likely benign.
