The p.A1130T variant (also known as c.3388G>A), located in coding exon 20 of the PTCH1 gene, results from a G to A substitution at nucleotide position 3388. The alanine at codon 1130 is replaced by threonine, an amino acid with similar properties. This alteration has been reported as a variant of unknown significance in an individual whose personal/family history met Bethesda criteria for Lynch syndrome (Martin-Morales L et al. PLoS One, 2018 Sep;13:e0203885). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
