The c.1503G>A variant in the PTCH1 gene has been previously reported in at least one individual pursuing genetic testing for Gorlin syndrome (Klein et al., 2005). Several in silico splice prediction models predict that c.1503G>A destroys the natural splice donor site in intron 10, and is expected to cause abnormal gene splicing. However, in the absence of RNA/functional studies, the actual effect of this sequence change is unknown. Based on currently available evidence, c.1503G>A is a strong candidate for a pathogenic variant. However, the possibility it is a rare benign variant cannot be excluded. -
Gorlin syndrome Uncertain:1
This sequence change affects codon 501 of the PTCH1 mRNA. It is a 'silent' change, meaning that it does not change the encoded amino acid sequence of the PTCH1 protein. This variant also falls at the last nucleotide of exon 10, which is part of the consensus splice site for this exon. This variant is not present in population databases (gnomAD no frequency). This variant has been observed in individual(s) with clinical features of nevoid basal cell carcinoma syndrome (PMID: 16301862). ClinVar contains an entry for this variant (Variation ID: 426225). Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
