In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies have shown that this missense change affects PCDH19 function (PMID: 34082468). This variant has not been reported in the literature in individuals affected with PCDH19-related conditions. This variant is present in population databases (rs772837341, gnomAD 0.004%). This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 198 of the PCDH19 protein (p.Arg198His). -
