Genes affected
PCDH19 (HGNC:14270): (protocadherin 19) The protein encoded by this gene is a member of the delta-2 protocadherin subclass of the cadherin superfamily. The encoded protein is thought to be a calcium-dependent cell-adhesion protein that is primarily expressed in the brain. Mutations in this gene on human chromosome X are associated with sporadic infantile epileptic encephalopathy and to a female-restricted form of epilepsy (EFMR; also known as PCDH19RE). Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2017]

The p.Y166* pathogenic mutation (also known as c.498C>A), located in coding exon 1 of the PCDH19 gene, results from a C to A substitution at nucleotide position 498. This changes the amino acid from a tyrosine to a stop codon within coding exon 1. p.Y166* resulting from c.497dupA (reported as c.497_498insA) has been identified in a Japanese female with epilepsy (Higurashi N et al. Epilepsy Res., 2013 Sep;106:191-9). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
