This variant is reported in ClinVar (Variation ID: 11381), and is absent from general population databases (Exome Variant Server, Genome Aggregation Database), indicating it is not a common polymorphism. The leucine at codon 542 is highly conserved, and computational analyses (SIFT, PolyPhen-2) predict that this variant is deleterious. Structural analyses of the BTK protein show that leucine 542 may be important for substrate binding (Mattsson 1996). However, given the limited amount of clinical data and lack of functional data, the significance of the p.Leu542Pro variant is uncertain at this time. References: Conley et al.
