Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 183636 control chromosomes (gnomAD). c.776C>G has been reported in the literature in multiple individuals affected with Fabry disease (Ashley_GLA_JHG_2001, Blaydon_GLA_Human_Mutation_2001, Shabbeer_GLA_HG_2006). Moreover, Shin et al. measured the GLA activity in two male Fabry disease patients who had the variant of interest and found that the enzyme activity was below 10% of the normal enzyme activity indicating a deleterious impact of the variant on the functionality of the protein. These data indicate that the variant is very likely to be associated with disease. The following publications have been ascertained in the context of this evaluation (PMID: 18698230, 16595074, 11668641, 11322659, 19343533, 23474038). This missense change has been observed in individuals with Fabry disease and hypertrophic cardiomyopathy (PMID: 11322659, 11668641, 16595074, 24582695, 27532257, 28756410). This variant is not present in population databases (gnomAD no frequency). This sequence change replaces proline, which is neutral and non-polar, with arginine, which is basic and polar, at codon 259 of the GLA protein (p.Pro259Arg). - |
