Sulfatases are essential for the correct composition of bone and cartilage matrix. X-linked chondrodysplasia punctata, a disease characterized by abnormalities in cartilage and bone development, has been linked to mutations in this gene. This sequence change replaces histidine, which is basic and polar, with leucine, which is neutral and non-polar, at codon 548 of the ARSE protein (p.His548Leu). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with ARSE-related conditions. ClinVar contains an entry for this variant (Variation ID: 1683177). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
