Deletions, duplications, and point mutations at this gene locus may cause Duchenne muscular dystrophy (DMD), Becker muscular dystrophy (BMD), or cardiomyopathy. This variant has been observed to be de novo in an individual affected with clinical features consistent with dystrophinopathy (Invitae). This variant is not present in population databases (ExAC no frequency). This sequence change replaces arginine with proline at codon 3370 of the DMD protein (p.Arg3370Pro). The arginine residue is highly conserved and there is a moderate physicochemical difference between arginine and proline. -
