This protein, which is a member of the peptidase C1 protein family, is predominantly expressed in osteoclasts. However, the encoded protein is also expressed in a significant fraction of human breast cancers, where it could contribute to tumor invasiveness. This variant disrupts a region of the CTSK protein in which other variant(s) (p.Cys318Tyr) have been determined to be pathogenic (PMID: 20814951, 24057333, 27558267). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with CTSK-related conditions. - |
