Genes affected
NR0B2 (HGNC:7961): (nuclear receptor subfamily 0 group B member 2) The protein encoded by this gene is an unusual orphan receptor that contains a putative ligand-binding domain but lacks a conventional DNA-binding domain. The gene product is a member of the nuclear hormone receptor family, a group of transcription factors regulated by small hydrophobic hormones, a subset of which do not have known ligands and are referred to as orphan nuclear receptors. The protein has been shown to interact with retinoid and thyroid hormone receptors, inhibiting their ligand-dependent transcriptional activation. In addition, interaction with estrogen receptors has been demonstrated, leading to inhibition of function. Studies suggest that the protein represses nuclear hormone receptor-mediated transactivation via two separate steps: competition with coactivators and the direct effects of its transcriptional repressor function. [provided by RefSeq, Jul 2008]
NUDC (HGNC:8045): (nuclear distribution C, dynein complex regulator) This gene encodes a nuclear distribution protein that plays an essential role in mitosis and cytokinesis. The encoded protein is involved in spindle formation during mitosis and in microtubule organization during cytokinesis. Pseudogenes of this gene are found on chromosome 2. NR0B2-related disorder Uncertain:1
Uncertain significance, no assertion criteria providedclinical testingPreventionGenetics, part of Exact SciencesAug 06, 2024The NR0B2 c.749T>C variant is predicted to result in the amino acid substitution p.Leu250Pro. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.017% of alleles in individuals of Latino descent in gnomAD. At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
Inherited obesity Uncertain:1
Uncertain significance, criteria provided, single submitterclinical testingNew York Genome CenterJul 13, 2023The c.749T>C p.(Leu250Pro) variant identified in the NR0B2 gene has not previously been reported in ClinVar or in any patient in the literature. It is observed in 9 alleles in population databases (~0.002% MAF with 0 homozytoe in gnomAD v2.1.1 and v3.1.2, TOPMed Freeze 8), suggesting that it is not a common benign variant in the populations represented in that database. The c.749T>C variant is located in exon 2 of this 2-exon gene and is predicted to replace a leucine amino acid with proline at codon 250 of the NR0B2 protein [PMID: 20970497]. In silico predictions are in favor of the p.(Tyr166Cys) variant's effect [REVEL = 0.897]; however, there are no functional studies to support or refute these predictions. Based on available evidence this c.749T>C p.(Leu250Pro) variant identified in NR0B2 is classified as a Variant of Uncertain Significance. -
not provided Uncertain:1
Uncertain significance, criteria provided, single submitterclinical testingLabcorp Genetics (formerly Invitae), LabcorpOct 22, 2023This sequence change replaces leucine, which is neutral and non-polar, with proline, which is neutral and non-polar, at codon 250 of the NR0B2 protein (p.Leu250Pro). This variant is present in population databases (no rsID available, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with NR0B2-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt NR0B2 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
