This protein interacts with nephrocystin, and belongs to a multifunctional complex that is localized to actin- and microtubule-based structures. Mutations in this gene are associated with nephronophthisis type 4, a renal disease, and with Senior-Loken syndrome type 4, a combination of nephronophthisis and retinitis pigmentosa. This variant has not been reported in the literature in individuals affected with NPHP4-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces isoleucine with serine at codon 1406 of the NPHP4 protein (p.Ile1406Ser). The isoleucine residue is highly conserved and there is a large physicochemical difference between isoleucine and serine. -
