Le ucine (Leu) at position 1003 is not conserved in mammals or evolutionarily dista nt species and 3 species (parrot, scarlet macaw, and painted turtle) carry a phe nylalanine (Phe) at this position, raising the possibility that this change may be tolerated. Additional computational prediction tools suggest that the variant may not impact the protein, though this information is not predictive enough to rule out pathogenicity. In summary, while the clinical significance of the p.Le u1003Phe variant is uncertain, these data suggest that it is more likely to be b enign. - |
