Uncertain significance, criteria provided, single submitter | clinical testing | All of Us Research Program, National Institutes of Health | Oct 06, 2023 | This missense variant replaces asparagine with serine at codon 2145 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in two individuals affected with breast cancer (PMID: 33471991; Leiden Open Variation Database DB-ID BRCA2_003373). This variant has been identified in 1/226590 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. - |

Likely benign, criteria provided, single submitter | clinical testing | Myriad Genetics, Inc. | Apr 02, 2024 | This variant is considered likely benign. This variant is strongly associated with less severe personal and family histories of cancer, typical for individuals without pathogenic variants in this gene [PMID: 25085752]. - |

Hereditary cancer-predisposing syndrome Uncertain:2Benign:1
Likely benign, criteria provided, single submitter | curation | University of Washington Department of Laboratory Medicine, University of Washington | Mar 23, 2023 | Missense variant in a coldspot region where missense variants are very unlikely to be pathogenic (PMID:31911673). - |
Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Dec 18, 2023 | The p.N2145S variant (also known as c.6434A>G), located in coding exon 10 of the BRCA2 gene, results from an A to G substitution at nucleotide position 6434. The asparagine at codon 2145 is replaced by serine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
Uncertain significance, criteria provided, single submitter | clinical testing | Color Diagnostics, LLC DBA Color Health | Jun 22, 2023 | This missense variant replaces asparagine with serine at codon 2145 of the BRCA2 protein. Uncertain significance, criteria provided, single submitter | clinical testing | GeneDx | Dec 22, 2014 | This variant is denoted BRCA2 c.6434A>G at the cDNA level, p.Asn2145Ser (N2145S) at the protein level, and results in the change of an Asparagine to a Serine (AAT>AGT). This variant, also known as c.6662A>G using alternate nomenclature, has not, to our knowledge, been published in the literature as pathogenic or benign. BRCA2 Asn2145Ser was not observed at a significant allele frequency in the NHLBI Exome Sequencing Project. Since Asparagine and Serine share similar properties, this is considered a conservative amino acid substitution. BRCA2 Asn2145Ser occurs at a position that is moderately conserved across species and is not located in a known functional domain. In silico analyses predict that this variant is unlikely to alter protein structure or function. Based on currently available information, it is unclear whether BRCA2 Asn2145Ser is pathogenic or benign. We consider it to be a variant of uncertain significance. - |
Hereditary breast ovarian cancer syndrome Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Nov 24, 2023 | This sequence change replaces asparagine, which is neutral and polar, with serine, which is neutral and polar, at codon 2145 of the BRCA2 protein (p.Asn2145Ser). This variant is present in population databases (rs80358878, gnomAD 0.0009%). This missense change has been observed in individual(s) with BRCA2-related conditions (PMID: 10923033). ClinVar contains an entry for this variant (Variation ID: 52094). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt BRCA2 protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. - |
