The glutamine at codon 133 is replaced by glutamic acid, an amino acid with highly similar properties. This alteration has been identified in multiple individuals diagnosed with breast cancer (Tung N et al. Cancer, 2015 Jan;121:25-33; Purrington KS et al. Cancer Epidemiol Biomarkers Prev, 2020 Nov;29:2369-2375; Dorling et al. N Engl J Med, 2021 02;384:428-439). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
