The variant was identified in dbSNP (ID: rs1018509561) and in control databases in 5 of 168452 chromosomes at a frequency of 0.00002968 (Genome Aggregation Database March 6, 2019, v2.1.1). The variant was observed in the following populations: Latino in 2 of 25310 chromosomes (freq: 0.000079), African in 1 of 15152 chromosomes (freq: 0.000066) and European (non-Finnish) in 2 of 69138 chromosomes (freq: 0.000029), but was not observed in the Ashkenazi Jewish, East Asian, European (Finnish), Other, or South Asian populations. The p.Ala7Val residue is not conserved in mammals and computational analyses (PolyPhen-2, SIFT, AlignGVGD, BLOSUM, MutationTaster) do not suggest a high likelihood of impact to the protein; however, this information is not predictive enough to rule out pathogenicity.
