The encoded protein is a pattern recognition receptor that detects cytosolic nucleic acids and transmits signals that activate type I interferon responses. The encoded protein has also been shown to play a role in apoptotic signaling by associating with type II major histocompatibility complex. This variant is not present in population databases (ExAC no frequency). This sequence change results in a premature translational stop signal in the TMEM173 gene (p.Thr342Metfs*5). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 38 amino acids of the TMEM173 protein. Experimental studies and prediction algorithms are not available for this variant, and the functional significance of the affected amino acids is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. - |
