Other variant(s) that disrupt this residue have been observed in individuals with PTEN-related conditions (PMID: 12372056, 23470840), which suggests that this may be a clinically significant amino acid residue. Experimental studies have shown that this missense change affects PTEN function (PMID: 32350270). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Not Available"). This missense change has been observed in individual(s) with macrocephaly (PMID: 23124040). This variant is not present in population databases (gnomAD no frequency). This sequence change replaces methionine, which is neutral and non-polar, with isoleucine, which is neutral and non-polar, at codon 134 of the PTEN protein (p.Met134Ile). - |
