This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. Experimental studies have shown that this missense change affects SLC4A11 function (PMID: 16767101, 29327391). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. This missense change has been observed in individuals with congenital hereditary endothelial dystrophy (PMID: 16767101, 17679935, 31714402). This variant is present in population databases (rs121909388, gnomAD 0.008%). This sequence change replaces serine, which is neutral and polar, with leucine, which is neutral and non-polar, at codon 489 of the SLC4A11 protein (p.Ser489Leu). -
