PVS1, PM2_Supporting c.1474-1G>T, located in a canonic splicing site of the BRIP1, is predicted to alter splicing, probably causing the skipping of exon 11 (r.1474_1628del). This alteration is expected to result in loss of function by premature protein truncation and nonsense-mediated mRNA decay (PVS1). It is not present in the population database gnomAD v2.1.1, non cancer dataset (PM2_supporting). To our knowledge, neither relevant clinical data nor well-stablished functional studies have been reported for this variant. Also, the variant has not been reported neither in ClinVar nor in LOVD databases. Based on the currently available information, c.1474-1G>T is classified as a likely pathogenic variant according to ACMG guidelines. -
