This premature translational stop signal has been observed in individual(s) with autosomal recessive congenital myopathy (PMID: 20418530). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is present in population databases (rs762121316, gnomAD 0.006%). This sequence change creates a premature translational stop signal (p.Arg783*) in the MYH2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MYH2 are known to be pathogenic (PMID: 20418530, 23388406, 24193343). -
