The APC c.4334C>T (p.Thr1445Ile) variant has been reported in the published literature in an individual with colorectal cancer whose tumor showed microsatellite stability with normal mismatch repair protein expression (PMID: 29458332 (2018)). In addition, the variant has been detected in tumors of an individual with oropharynx cancer (PMID: 25742471 (2015)), and an individual with sebaceous adenoma (PMID: 27311873 (2016)). The frequency of this variant in the general population, 0.000016 (4/251168 chromosomes (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is benign. Based on the available information, we are unable to determine the clinical significance of this variant. -
