The BRIP1 c.290_293del (p.Asn97Metfs*3) variant alters the translational reading frame of the BRIP1 mRNA and causes the premature termination of BRIP1 protein synthesis. This variant has been reported in the published literature individuals with ovarian cancer (PMID: 31341520 (2018)) and serous cystadenocarcinoma (PMID: 36451132 (2022)). This variant has also been reported in unaffected individuals (PMID: 33471991 (2021), see also LOVD (http://databases.lovd.nl/shared/genes/BRIP1)). Based on the available information, this variant is classified as pathogenic. The frequency of this variant in the general population, 0.000008 (2/251452 chromosomes (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is consistent with pathogenicity. -

The c.290_293delACAA pathogenic mutation, located in coding exon 3 of the BRIP1 gene, results from a deletion of 4 nucleotides at nucleotide positions 290 to 293, causing a translational frameshift with a predicted alternate stop codon (p.N97Mfs*3). This alteration has been reported in multiple ovarian cancer patients (Kanchi KL et al. Nat Commun, 2014;5:3156; Lu C et al. Nat Commun. 2015 Dec 22;6:10086; Arvai KJ et al. Hered Cancer Clin Pract, 2019 Jul;17:19). One study detected this mutation in 0/3030 pancreatic cancer cases and 2/123136 population controls (Hu C et al. JAMA, 2018 06;319:2401-2409). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
