The p.I183T variant (also known as c.548T>C), located in coding exon 3 of the RAD51C gene, results from a T to C substitution at nucleotide position 548. The isoleucine at codon 183 is replaced by threonine, an amino acid with similar properties. In one study, this variant was seen in 1 of 1404 breast and/or ovarian cancer cases with family histories of breast and/or ovarian cancer and in 0 of 1156 population controls (Loveday C et al. Nat. Genet. 2012 Apr;44:475-6; author reply 476). In a homology-directed DNA repair (HDR) assay, this alteration showed a functionally normal read-out (Hu C et al. Cancer Res, 2023 Aug;83:2557-2571). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This missense variant replaces isoleucine with threonine at codon 183 of the RAD51C protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been observed in an individual affected with ovarian and/or breast cancer (PMID: 22538716). This variant has been identified in 2/251468 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

This variant is present in population databases (rs756727559, gnomAD 0.002%). This missense change has been observed in individual(s) with ovarian cancer (PMID: 22538716). ClinVar contains an entry for this variant (Variation ID: 187692). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Experimental studies have shown that this missense change does not substantially affect RAD51C function (PMID: 37253112). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
