The isoleucine at codon 284 is replaced by threonine, an amino acid with similar properties. This variant has been reported in at least two individuals with hypertrophic cardiomyopathy (Robyns T et al. Eur J Hum Genet, 2017 12;25:1313-1323; Robyns T et al. Eur J Med Genet, 2020 Mar;63:103754). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive.
