This variant has also been reported in ClinVar (Variation ID#:189099) and has been interpreted as pathogenic by Invitae and Natera, Inc, and likely pathogenic by Counsyl. This frameshift variant leads to a premature termination codon at amino acid position 338 which is predicted to lead to a truncated or absent protein. Loss of function is an established disease mechanism for POMGNT1-associated muscular dystrophy-dystroglycanopathy. In summary, the clinical significance of the p.Asp338Ter variant is uncertain.
